In May, I completed my undergraduate thesis project at the Workenhe Lab. These past nine months have taught me invaluable lessons in research conceptualization, from experimental design and logistical planning to the communication of results. Each step along this journey has equipped me with skills that will undoubtedly serve me in the future. In the latter half of my undergraduate thesis project, I accomplished the following: 🔬 Established homologous recombination donor plasmids for HSV-1 engineering using established cloning techniques. 🔬 Designed and executed an experiment involving mammalian cell transfection, precise timing of trigger molecule addition, fluorescent microscopy, and immunoblotting to demonstrate successful inducible expression of the gene of interest in vitro. 🔬 Demonstrated robust cell culture maintenance and inventory management skills by successfully propagating MDCK, CRFK, and RK13 cell cultures for herpes virus infection. 🔬 Exhibited diligence and integrity in establishing cell line-specific and virus-specific cell culture basics (DMEM, PBS, TrypLE) to prevent contamination. 🔬 Presented research progress and findings to peers and faculty in the form of a poster presentation. 🔬 Communicated the research question, findings, and discussion of findings of the undergraduate project in a scientific manuscript. Cell division in your body unfolds in several sequential steps:                                    🔹 DNA Replication: The cell duplicates its DNA to ensure both new cells inherit identical genetic material. 🔹 Chromosome Formation: The copied DNA condenses into structures known as chromosomes. 🔹 Chromosome Alignment: These chromosomes align at the cell's center, preparing for separation. 🔹 Chromosome Separation: The cell pulls the chromosomes apart, ensuring an even distribution between two regions. 🔹 Cell Division: Finally, the cell undergoes cytokinesis, pinching in the middle to form two separate cells. PhD studentship opportunity: Probing the Function of RNA Structural Elements in the FMDV Genome using Biophysical and Genetic Approaches. Foot-and-mouth disease (FMD) is a highly contagious disease of cattle, sheep and pigs that causes huge economic loss in some of the poorest countries in the world and poses to significant threat to food security. This collaborative project with the University of York will investigate RNA structures essential for FMDV replication. 15 PhD fellowships in molecular microbiology, over molecular and structural biology (using NMR, X-ray crystallography, and cryogenic EM), and computational, organic and medicinal chemistry to assay development, fragment and high-throughput screening. In recent years it became increasingly evident that RNAs constitute promising macromolecular drug targets for small molecule drugs. By targeting upstream RNA, current undruggable protein-mediated pathways could be effectively modulated thereby opening for innovative therapies that may redefine the treatment strategies for a range of conditions. The TargetRNA doctoral training network brings together expertise and methods from molecular biology, structural biology, biophysics, computational chemistry, synthetic and medicinal chemistry, and microbiology, all the way up to in vivo models to discover new small molecule RNA ligands for selected targets and to characterize the effects of these RN A ligands on microbes and the microbiome. The network will train the doctoral candidates (DCs) with cutting-edge interdisciplinary skills in the discovery and characterization of RNA ligands and the translation of these discoveries into innovations.
